Essential Job Duties
This post doctoral scholar position will primarily be engaged in research into mast cell biology and signaling pathways involved in mast cell-mediated diseases including mastocytosis and allergic diseases. In addition, the post doc will carry out research into lung structural cell and immune cell interactions and signaling pathways involved in airway physiology. The post doc must be able to work independently, but must also be able to integrate into a multidisciplinary research environment that includes multiple research groups and collaborations in bioengineering and chemistry.

Duties and Responsibilities The Cruse lab is seeking a postdoctoral scholar to join the team and carry out research projects on examining mast cell signaling and molecular biology. The goal of our research program is to establish novel regulators of mast cell signaling and function, and to develop novel targeted therapeutics for mast cell-mediated diseases. The laboratory has expertise in in vitro, ex vivo and in vivo approaches of studying molecular immunology, cell signaling and mast cell biology in inflammation and mastocytosis. We utilize molecular and translational medicine approaches to identify potential innovative therapeutics that target mast cell function and allergic inflammation, as well as mast cell neoplasms. Overall, we strive to better understand the molecular mechanisms underlying mast cell-mediated diseases. We have a particular interest in identifying associations between proteins and genes with asthma pathogenesis and identifying the functions of novel gene candidates in human lung and cord blood-derived cells. We are achieving these goals through interdisciplinary collaborations with both clinical and basic scientists in various related fields.
The incumbent independently performs research on human and murine mast cells and other immune cells through the utilization of in vitro molecular and cell biology methods. The incumbent will also perform cell isolations and cultures from tissue, including lung tissue, and perform in vivo models of allergic inflammation and mastocytosis (mice models).
Oversee and maintain the lab tissue culture and maintain ongoing cell cultures of both primary cells and cell lines. Isolate human cells from primary tissues and blood products. Independently design, perform and analyze cellular and molecular biology experiments. Independently design, perform and analyze research on human, murine and canine mast cells through the utilization of cell cultures and in vivo models (mice models). Isolate human mast cells from lung tissue. Isolate and culture mast cells from blood. Develop research methods and generate research results that will contribute toward publications and presentations. Teach students to advance their understanding of the subject matter and add to/increase current laboratory procedures skills. Train visiting researchers, students and staff in biosafety and laboratory procedures. Performing work associated with research grants and patents, gathering preliminary data for grants and patents, writing and preparing manuscripts for publication, presenting research at conferences, assisting in graduate student training, ordering supplies and other laboratory operations.

Work Schedule Monday – Friday, 8am – 5pm; some weekends and holidays may be required.

Ph.D. or equivalent doctorate (e.g., M.D., D.V.M., Sc.D.) in appropriate field.
Preferred Years Experience, Skills, Training, Education
Extensive experience in cell biology, tissue/cell culture and aseptic technique are essential. Experience with isolation and culture of cells from tissues is essential. Experience in RT-PCR, cell transfection, cloning techniques, molecular biology and Western blots are preferred. Experience with mouse models preferred.
This position requires excellent writing and communications skills and the researcher is expected to publish their findings in acceptable peer-reviewed publications and present their work at national and international meetings.
Laboratory experience in human lung mast cell isolation and culture, primary human airway smooth muscle, fibroblast and airway epithelial cell culture (including differentiated ALI culture models), RNA isolation and quantification, quantitative PCR, protein extraction and quantification, ELISAs, Western blotting and immunohistochemistry on lung sections would be preferred.
